The active ingredient in senna leaves is called sennoside
. This molecule is converted by microorganisms in the colon into another substance, anthrone rhein, which has the beneficial effects of stimulating colonic activity (which accelerates intestinal motility and improves digestion) and increases fluid secretion. The laxative effects of sennoside and its active metabolite anthrone rhein are thought to inhibit the absorption of water and electrolytes in the large intestine, thereby increasing the volume and pressure of intestinal contents. It will stimulate the propulsive tightening movement of the colon. Additionally, stimulation of reactive chloride secretion increases intestinal water and electrolyte content. These changes in active electrolyte transport are dependent on calcium at the serosal surface. The laxative effect of sennosides is partly through stimulation of colonic fluid and electrolyte secretion, and this secretory capacity is regulated by stimulation of the formation of endogenous prostaglandins. Sennosides may be prepared as enemas or suppositories or mixed with stool softeners or fiber-blocking laxatives to form combined laxatives.
